چکیده

Our laboratory has characterized androgen metabolism in an androgen-responsive prostate cancer cell line (LNCaP) and showed that these cells accumulated intracellular testosterone primarily as glucuronidated metabolites. Using a cell-free assay with testosterone as substrate, we showed that LNCaP had UDP-glucuronosyltransferase (UDPGT) activity. Because dietary factors, such as flavonoids in soy products, may reduce the risk for hormone-dependent cancers, we studied the effects of fla vonoids on testosterone-UDPGT activity. LNCaP cells were exposed to selected flavonoids for up to 6 days. The increase in UDPGT-specific activity was linear over this period. Of the compounds tested, biochanin A was the most potent, with increased activity at concentration range 0.5-50 JIM. Activities were linear for time and protein and were unaffected by flavonoids added directly to the assay. Kinetics studies showed no change in A,,, for testosterone in the face of these large increases in specific activity. Cellular metabolism of testosterone reflected the increase in enzyme activity. Intact cells treated with biochanin A produced testosterone-glucuronide from testosterone at twice the rate of controls. The steroid form of the UDPGT transcript was expressed in LNCaP cells and was enhanced in biochanin A-treated LNCaP cells. Additionally, bio chanin A markedly decreased prostate specific antigen (PSA) level against the effect of testosterone on PSA production. Biochanin A significantly decreased the testosterone-stimulated release of PSA, presumably because biochanin A increased UDPGT and increased the intracellular glucuronidation of testosterone. These studies suggest that the modulation of hormone metabolism by dietary factors may be important in the preven tion and treatment of prostate cancer.

Engineered Jurkat Cells for Targeting Prostate-Specific Membrane Antigen on Prostate Cancer Cells by Nanobody-Based Chimeric Antigen Receptor

Background: Recently, modification of T cells with chimeric antigen receptor (CAR) has been an attractive approach for adoptive immunotherapy of cancers. Typically, CARs contain a single-chain variable domain fragment (scFv). Most often, scfvs are derived from a monoclonal antibody of murine origin and may be a trigger for host immune system that leads to the T-cell clearance. Nanobody is a spe...

Prostate specific antigen staining intensity in prostatic adenocarcinoma correlates with serum prostate specific antigen , Bandar Abbas, Iran

Introduction: Prostatic adenocarcinoma is one the most common malignances and the second cause of death in males due to cancer. Prostate Specific Antigen (PZSA) serum level is the first marker to screen such patients. We aimed in this study, to assess the intensity of prostatic adenocarcinoma tissue staining with PSA staining method. Methods: In this corss-sectional study, prostatic adenocarcin...

Can Prostate Specific Antigen Be Used as New Biomarker for Early Diagnosis of Breast Cancer?

Introduction: As a glycoprotein, Plasma Prostate-Specific Antigen (PSA) is mainly produced by prostate epithelial cells and is used as a major diagnostic tool for prostate cancer. A group of researchers relate the elevated number of estrogen receptors in breast cancer samples to the over-production of PSA in this type of cancer. Therefore, the present study aimed to determine the participants' ...

PRODUCTIO N OF PROSTATE-SPECIFIC ANTIGEN (PSA) BY A BREAST CANCER CELL LINE, SK-Br-3

PSA is a 33-KDa serine protease that is produced predominantly by prostate epithelium. However, it has been shown that about 30-40% of female breast tumors produce PSA and its production is associated with the presence of estrogen and progesterone receptors. We have now developed a new tissue culture system to study PSA production in breast cancer and its association with prognostic factors...
